OpenVent DNA polymerase

Description

OpenVent is an off-patent version of a thermostable archaeal family-B DNA polymerase from Pyrococcus strain GB-D which is sold under the trademark DeepVent by NEB.

It has 5X higher fidelity than Taq and extremely high thermostability (half-life of 23 hours at 95°C). It is ideal for GC-rich, looped sequence and other difficult templates.

We have a construct for production of  OpenVent, which was designed by collaborators from the Reclone network. It is codon-optimised for expression in E.coli and is very robust and amenable to most forms of expression and purification.

Protocol summary

The plasmid is transformed to an  E. coli  strain for T7 expression system, typically BL21(DE3), for protein expression.

Purification is performed using Ni-NTA agarose and Heparin chromatography.
